NCIt definition : A family of basic helix-loop-helix heterodimeric transcription factors that activate
gene expression when oxygen levels decrease in the cellular environment. Each HIF
complex is comprised of an alpha subunit and beta subunit, which are encoded by separate
genes.;
NCIt note : During normoxic conditions, the alpha subunits of HIF are hydroxylated at conserved
proline residues by prolyl-hydroxylase enzymes. This modification promotes alpha subunit
ubiquitination, which leads to proteasomal degradation. In hypoxic conditions, prolyl-hydroxylase
activity is inhibited; therefore, the half-life of the alpha subunits are prolonged
and they are able to dimerize with the appropriate beta subunit to regulate the expression
of hypoxia-responsive genes.;
