NCIt definition : Poly(ADP-ribose) glycohydrolase (976 aa, 111 kDa) is encoded by the human PARG gene.
This protein is involved in hydrolysis of the ribose-ribose bonds present in poly(ADP-ribose).;
NCIt note : Poly(ADP-ribose) glycohydrolase acts both as an endo- and exoglycosidase, yielding
both ADP-ribose monomers and poly(ADP-ribose) chains of various lengths. However,
this enzyme is not able to cleave the ester bond between the terminal ADP-ribose and
ADP-ribosylated amino acid residues, yielding mono-ADP-ribosylated proteins. (UniProt); Poly(ADP-ribose) glycohydrolase protein is expressed as at least 5 isoforms produced
by alternative splicing. The isoforms are named by their apparent molecular weight:
isoform 1 (the UniProt canonical isoform) is hPARG111; isoform 2 hPARG102 is missing
amino acid residues 1-89; isoform 3 hPARG99 is missing amino acid residues 1-108;
isoform 4 hPARG60 is catalytically inactive and has variant amino acids for residues
1-15, is missing residues 16-423 and has a substitution where amino acid residues
485-526 are replaced by tryptophan; and isoform 5 hPARG55 is also catalytically inactive
and is missing amino acid residues 1-460 and has a substitution where amino acid residues
485-526 are replaced by tryptophan. (UniProt);
