NCIt definition : Paired box protein PAX-2 (417 aa, 45 kD) is encoded by the human PAX2 gene. This
soluble, nuclear protein plays a role in transcription by RNA polymerase II and cellular
proliferation.;
NCIt note : Presence of PAX2 in tumor cells blocks production of the ERBB2 protein, which fuels
tumor growth. An absence or deficiency of PAX2 decreases the effectiveness of tamoxifen.
Several new DNA binding sites for Estrogen Receptor were found in the ERBB2 gene,
meaning that liganded ER is able to upregulate its own gene. When exposed to tamoxifen,
PAX2 binds to ER and ablates this upregulation. The absence of PAX2 may be used to
identify patients who will not respond to to tamoxifen, but will respond to alternate
