NCIt definition : Human GTF2B wild-type allele is located within 1p22-p21 and is approximately 39 kb
in length. This allele, which encodes transcription initiation factor IIB protein,
is involved in transcription through the modulation of the formation of the RNA polymerase
II preinitiation complex.;
NCIt note : The GTF2B (TFIIB) gene encodes a protein that is a general transcription factor and
plays a central role in preinitiation complex assembly. TFIIB is one of the ubiquitous
factors required for transcription initiation by RNA polymerase II. TFIIB localizes
to the nucleus where it forms a complex (the DAB complex) with transcription factors
IID and IIA. TFIIB provides a bridge between promoter-bound TFIID and RNA Polymerase
II. TFIIB possesses sequence-specific DNA-binding ability and interacts with the TFIIB-recognition
element (BRE), present in many promoters. BRE suppresses the basal level of transcription
elicited by a core promoter, which increases the amplitude of transcriptional stimulation
in the presence of an activator where an activator can disrupt the TFIIB-BRE interaction
within a promoter-bound complex. The TFIIB gene is located at chromosome 1p22-p21
on the short arm of chromosome 1. (from PMID 11711430, LocusLink 2959);
