NCIt definition : A preparation of allogeneic human Epstein-Barr virus (EBV)-sensitized T-lymphocytes
that have been engineered to express a chimeric antigen receptor (CAR) specific for
the tumor-associated antigen (TAA) cluster of differentiation 19 (CD19), with a modified
CD3zeta signaling domain 1xx, with potential immunostimulating and antineoplastic
activities. Upon administration, allogeneic anti-CD19 CAR T-cells ATA3219 recognize
and bind to CD19-expressing tumor cells, thereby inducing selective toxicity in CD19-expressing
tumor cells. CD19 antigen is a B-cell specific cell surface antigen expressed in all
B-cell lineage malignancies. The use of allogeneic EBV T-cells minimizes the risks
for graft-versus-host disease (GvHD). The modified CD3zeta signaling domain 1xx retains
signaling capacity in 1 of 3 immune-receptor-tyrosine-based-activation-motif (ITAM)
regions, which may help prevent counterproductive T-cell differentiation and exhaustion,
and may enhance the anti-tumor activity of the CAR T-cells.;
