NCIt definition : A preparation of human allogeneic CD4 and CD8 T-lymphocytes transduced with a chimeric antigen receptor (CAR) specific for the tumor-associated antigen (TAA) CD22, and ex vivo modified to disrupt the expression of major histocompatibility (MHC) class I and MHC class II molecules and to express CD47, with potential immunomodulating and antineoplastic activities. Upon introduction into the patient, the allogeneic hypoimmune anti-CD22 CAR T-cells SC262 recognize and bind to CD22-expressing tumor cells. This may result in a specific cytotoxic T-lymphocyte (CTL)-mediated killing of CD22-positive tumor cells. CD22, a B-lineage-restricted, transmembrane phosphoglycoprotein, is expressed on malignant B-cells. The disruption of MHC class I and MHC class II molecules and the expression of CD47 prevent both innate and adaptive immune responses against the allogeneic CAR T-cells, thereby increasing the persistence of the CAR T-cells.;
