NCIt definition : A preparation of human allogeneic T-lymphocytes transduced with a chimeric antigen
receptor (CAR) specific for the tumor-associated antigen (TAA) CD19, and modified
ex vivo to disrupt the expression of major histocompatibility (MHC) class I and MHC
class II molecules and to express CD47, with potential immunomodulating and antineoplastic
activities. Upon introduction into the patient, the allogeneic hypoimmune anti-CD19
CAR T-cells SC291 recognize and bind to CD19-expressing tumor cells. This may result
in a specific cytotoxic T-lymphocyte (CTL)-mediated killing of CD19-positive tumor
cells. CD19 antigen is a B-cell specific cell surface antigen expressed in all B-cell
lineage malignancies. The disruption of MHC class I and MHC class II molecules and
the expression of CD47 prevent immune response against the allogeneic CAR T-cells,
thereby increasing the persistence of the CAR T-cells.;
