NCIt definition : An antibody-drug conjugate (ADC) consisting of an anti-c-Met monoclonal antibody that is conjugated to monomethyl auristatin E (MMAE), an auristatin derivative and potent microtubule disrupting agent, with potential antineoplastic activity. Upon administration of anti-c-Met/MMAE ADC RC108, the monoclonal antibody moiety targets and binds to the c-Met protein, which is overexpressed in certain tumor types. After internalization of the agent, the MMAE moiety is released and binds to tubulin and inhibits its polymerization, which results in G2/M phase arrest and apoptosis. This inhibits the proliferation of c-Met-expressing tumor cells. c-Met, also known as hepatocyte growth factor receptor (HGFR), is a receptor tyrosine kinase that is overexpressed or mutated in many tumor cell types and plays a key role in tumor cell proliferation, survival, invasion, metastasis, and tumor angiogenesis. Its expression is associated with poor prognosis in many solid tumor types.;
