NCIt definition : An antibody-drug conjugate (ADC) composed of an antibody targeting CC chemokine receptor 7 (CCR7) and conjugated to the cytotoxic maytansinoid DM4, with potential antineoplastic activity. Upon administration of anti-CCR7 ADC JBH492, the antibody moiety targets and binds to CCR7 on tumor cells. Upon antibody/antigen binding and internalization, the ADC releases DM4, which binds to tubulin and disrupts microtubule assembly/disassembly dynamics. This results in the inhibition of cell division and cell growth of CCR7-expressing tumor cells. CCR7, a G-protein coupled receptor, is normally expressed by subsets of immune cells and overexpressed by various types of cancer cells. Its overexpression has been associated with lymph node metastasis and poor survival.;
