NCIt definition : Human DPYD wild-type allele is located within 1p22 and is approximately 843 kb in
length. This allele, which encodes dihydropyrimidine dehydrogenase [NADP ] protein,
plays a role in the reduction of uracil, thymine and the drug 5-fluorouracil. Hereditary
defects in the DPYD gene cause an error in pyrimidine metabolism associated with both
thymine-uraciluria and increased toxicity of 5-fluorouracil during chemotherapy.;
NCIt note : Increased levels of dihydropyrimidine dehydrogenase [NADP ] alter the efficacy of
5-fluorouracil-based chemotherapy via increased inactivation of this agent in tumor
cells. Levels of DPYD mRNA in cancerous tissues seem to correlate with tumor progression.
In both liver metastasis and advanced colorectal cancer, high DPYD mRNA levels may
be clinically important with respect to 5-fluorouracil-based chemosensitivity.;
