NCIt definition : A topical preparation containing the R100E mutated form of the human recombinant nerve growth factor (NGF), with potential neuroprotective, neurotrophic and wound healing activities and with reduced pain induction activity. Upon topical administration, mutated rhNGF CHF6467, by mimicking the natural-occuring neurotrophin, binds with high affinity to the tropomyosin receptor kinase A (TrkA) NGF receptor located on peripheral and central neurons and activates protein kinase B/mammalian target of rapamycin (mTOR) pathway signaling. This induces NGFs pleiotropic effects and may prevent prevent loss of nerve-endings and visual damage, and may promote sensory skin innervation, neoangiogenesis and skin wound healing. The R100 mutation abolishes the hyperalgesic effect that is usually associated with NGF without affecting most neurological functions.;
