targets the cell surface glycoprotein CD-38 and carries the E430G mutation, with potential
immunomodulating and antineoplastic activities. Upon administration, erzotabart targets
and binds to CD38 on CD38-positive tumor cells, thereby forming antibody hexamers
through increased intermolecular Fc-Fc interactions upon antigen binding. This may
trigger direct cell killing, antibody-dependent cellular cytotoxicity (ADCC), antibody-dependent
cell mediated phagocytosis (ADCP) and antibody-mediated complement dependent cytotoxicity
(CDC) in CD38-expressing tumor cells. In addition, binding to CD38 by GEN3014 induces
downmodulation of CD38, inhibits CD38 cyclase activity, and may abrogate immune suppression
in the tumor microenvironment (TME). CD38, a type II transmembrane glycoprotein, is
present on various immune cells and hematologic malignancies.;
