NCIt definition : Transcription factor p65 (551 aa, 60 kDa) is encoded by the human RELA gene. This
protein is involved in the modulation of both signaling and gene expression.;
NCIt note : NF-Kappa B p65 belongs to the Rel/Dorsal family. IKB kinases phosphorylate IkB and
activate NFKB via destructive ubiquitination. Translocated, activate NFKB transcription
factor binds the enhancer Kappa-B 5'-GGRNNYYCC-3' DNA motif, in immune response and
acute phase reaction genes. NF-Kappa B p65 (551-aa) is an NFKB subunit containing
rel homology regions, an alpha helical segment, and NLS that activates transcription
as a heterodimer with a 50-kd DNA-binding (kappa-B motif) subunit. Cytoplasmic NFKB,
sequestered by IKB interaction with p65, translocates to the nucleus on IKBKA/B-dependent
ubiquitinated destruction of IKB. p65 ser276 phosphorylation by PKA stimulates coactivator
CBP/p300 interaction and transcriptional activity. NFKB(pp65) associates with CBP
and displaces DNA bound p50-HDAC1 complexes that suppress gene expression. Through
N-terminal association, HDAC3 inhibits p65, abolishes CBP/p300 RELA acetylation, and
promotes XPO1-dependent RELA/IKBA binding for nuclear export. (from LocusLink, Swiss-Prot,
