NCIt definition : E3 ubiquitin-protein ligase Mdm2 (491 aa, 55 kDa) is encoded by the human MDM2 gene.
This protein is involved in the mediation of the ubiquitination and degradation of
protein substrates, and the inhibition of apoptosis induction that is mediated by
cellular tumor antigen p53.;
NCIt note : Inhibits p53 trans-activation, cell cycle arrest, and apoptosis. MDM2 binds p53 in
the cell cycle at S/G2M phase and targets p53 for proteasome-mediated proteolysis.
MDM2 also inhibits Rb protein and binds p14(ARF), ribosomal protein L5, and specifically
to RNA. MDM2 gene expression is induced by DNA damage and regulated by the Ras-MAP
kinase pathway. Some soft tissue sarcomas carry amplified MDM2 and overexpress MDM2
protein. (from SWISS-PROT Q00987, OMIM 164785, and NCI);
