from an antibody targeting the cell surface glycoprotein ADP-ribosyl cyclase 1 (CD38)
that is fused to a cytotoxic payload composed of the enzymatically active, de-immunized,
ribosome-inactivating cytotoxic payload Shiga-like toxin-A subunit (SLTA), with potential
antineoplastic activity. Upon administration of ETB targeting CD38 MT-0169, the anti-CD38
scFv moiety specifically targets and binds to CD38-expressing tumor cells. Upon internalization,
the SLTA moiety is released and acts as an N-glycosidase, which binds to and removes
an adenine nucleobase from the 28S RNA component of the 60S ribosomal subunit of ribosomes.
This irreversibly inhibits ribosome activity, which inhibits protein synthesis and
leads to apoptosis in CD38-expressing tumor cells. CD38, a type II transmembrane glycoprotein,
is present on various immune cells and hematologic malignancies; its expression has
been correlated with poor prognosis.;
